چکیده

3,4-Dihydroxybenzalacetone (DBL) is a polyphenol derived from the medicinal plant Chaga [Inonotus obliquus (persoon) Pilat]. Although Chaga is used in Russia folk medicine to treat tumors, very little is known about its mechanism of action. Because most genes involved in inflammation, antiapoptosis, and cell proliferation are regulated by the transcription factor nuclear factor-KB (NF-KB), we postulated that DBL activity is mediated via modulation of the NF-KB activation pathway. We investigated the effects of DBL on NF-KB activation by electrophoretic mobility shift assay and on NF-KB-regulated gene expression by Western blot analysis. We found that DBL suppressed NF-KB activation by a wide variety of inflammatory agents, including tumor necrosis factor (TNF), interleukin-1B, epidermal growth factor, okadaic acid, phorbol 12-myristate 13-acetate, and lipopolysaccharide. The suppression was not cell type specific and inhibited both inducible and constitutive NF-KB activation. DBL did not interfere with the binding of NF-KB to DNA but rather inhibited IKBA kinase activity, IKBA phosphorylation and degradation, p65 phosphorylation, and translocation. DBL also suppressed the expression of TNF-induced and NF-KB-regulated proliferative, antiapoptotic, and metastatic gene products. These effects correlated with enhancement of TNF-induced apoptosis and suppression of TNF-induced invasion. Together, our results indicate that DBL inhibits NF-KB activation and NF-KB-regulated gene expression, which may explain the ability of DBL to enhance apoptosis and inhibit invasion. N-(4-Hydroxyphenyl)Retinamide Inhibits Invasion, Suppresses Osteoclastogenesis, and Potentiates Apoptosis through Down-regulation of IKBA Kinase and Nuclear Factor-KB–Regulated Gene Products

N-(4-hydroxyphenyl) retinamide [4-HPR], a synthetic retinoid, has been shown to inhibit tumor cell growth, invasion, and metastasis by a mechanism that is not fully understood. Because the nuclear factor-KB (NF-KB) has also been shown to regulate proliferation, invasion, and metastasis of tumor cells, we postulated that 4-HPR modulates the activity of NF-KB. To test this postulate, we examined ...

Matrine inhibits diethylnitrosamine-induced HCC proliferation in rats through inducing apoptosis via p53, Bax-dependent caspase-3 activation pathway and down-regulating MLCK overexpression

The proliferation of hepatocellular carcinoma (HCC) cells is one of the leading causes of liver cancer mortality in humans. The inhibiting effects of matrine on HCC cell proliferation have been studied, but the mechanism of that inhibition has not been fully elucidated. Since, apoptosis plays an important role in HCC cell proliferation. We examined the apoptosis-inducing effect of matrine on tu...

Apoptosis Induction by a Novel Retinoid-Related Molecule Requires Nuclear Factor-KB Activation

Nuclear factor-KB (NF-KB) activation has been shown to be both antiapoptotic and proapoptotic depending on the stimulus and the specific cell type involved. NF-KB activation has also been shown to be essential for apoptosis induction by a number of agents. The novel retinoid-related molecule 4-[3Cl-(1-adamantyl)-4-hydroxyphenyl]-3-chlorocinnamic acid (3Cl-AHPC) activates NF-KB with subsequent a...

Coronarin D, a labdane diterpene, inhibits both constitutive and inducible nuclear factor-KB pathway activation, leading to potentiation of apoptosis, inhibition of invasion, and suppression of osteoclastogenesis

Compounds isolated from members of the Zingiberaceae family are traditionally used as a medicine against inflammatory diseases, but little is known about the mechanism. Here, we report the isolation and structural identification of coronarin D [E-labda-8(17),12-diene-15-ol], a labdane-type diterpene, from Hedychium coronarium and delineate its mechanism of action. Because the transcription fact...
